I have opened a box of nickels (Canadian) and how found many really nice looking ones from the 60s and 70s. Now I am trying to grade them. Some of them are certainly AU most are lower but is it possible some of my high AUs are actually low MS or is that impossible since they were in a roll?

It definitely is. MS60+ is determined by it's condition, not it's history. Theoretically you can find MS or even PF 70 coins in circulation. In practice I think it's not uncommon to find MS60-62 coins in circulation but much above that are rarities. I'm no expert though. Welcome to CRH'ing.

Can you point me to a comprehensive guide that shows the difference. I feel like I can grade circulated stuff no problem but I am lost at the bridge between circulated and non circulated

I use the PCGS photo grade website but I don't believe they have a similar site for Canadian coins. Sorry I can't help.
